It may be an infection that will cause her to go deaf. Call a vet and ask them.

It's an ear abcess, which needs to be drained. It is caused by improper humidity (often too dry). If you have medical training, I can explain how to perform the procedure or bring it to a qualified reptile vet. Here is a listof reptile vets by state:

P.S. Do this ASAP, it's not life threatening, but it is painful for the turtle.

Is the lump discolored at all? If it is, you might want to take it to a vet. It's some kind of disease I remember reading about, but I don't remember what it's called. I have a yellow bellied slider. It's a disease that affects turtles.

The best thing is take it to a vet, but it probably and abcess, I have had an aquatic turtle with that. It took a long time to heal.

www.boxturtles.com is a good web site to use for box turtle care. Also, www.billsboxturtles.com is another one I use every day.
You need to open the abcess and wash it with saline water, and put some antibiotic ointment on the wound.
If in doubt, please take it to a vet.
