If you are in the middle of Himayat Sagar (It is a big lake), then the best way is to first take a boat back to the shore and from the bus stop on the shore, you can get a direct a local city bus going to Charminar.
The other options are you can swim to reach the shore.
You can also take the same bus or three wheeler which takes you to the shores of Himayat Sagar.

from himayatsagar... get to mehdipatnam(u can do that using buses, seven seaters and also take a lift from college students)... then... from mehdipatnam take bus no: 65/66... u will reach charminar in half an hour depending on the traffic!1
