Depends on what interests you. Do you enjoy museums and such? Do you want to just hang out, have some picnics in the parks, stroll in the meditation gardens or temple grounds? Do you want to take a day trip to the Ajanta or Ellora caves (99 kms away)? First I would suggest thinking about your plans for the stopover -- what do you want to do with that time? If you have the time, I would suggest spending 2-3 days seeing Pune, and then maybe another day or two to see the caves nearby.
I disagree with the person who said there's not much to see in Pune. I have very fond memories of that city and can't wait for my next trip back.

Pune is a very small city you can easily visit all parts of the city within 2 days, the must watch destinations are Aga Khan palace and Abu garh

There is not much to c in Pune. The Aga Khan palace where Gandhi was imprisoned is a MUST. One day is enough.
