it is how steep the highway is if it says 6% that means the road will rise or drop 6 feet every 100 feet and on the U.S. Interstate system that is the steepest grade you will find it it put there as a warning to trucks (semi's) to let them know to down shift

its there for your safety but mostly for truck drivers pulling heavy loads it lets them know ahead of time to be in the correct gear to safely climb the hill or upgrade but they are also posted showing that there is a downgrade(going down the hill) so you can adjust your speed to not go over the speed limit that will cause the brakes on big trucks to get hot and not work,even catch on fire , then you will have a deadly accident.
