A taxi with a metered fare (plus tolls) will run approximately $40-55. There is an additional charge for luggage over 24 inches if the driver handles the bags, and rates are based per car, not per passenger.
But you also might want to consider Airtrain which can actually be much quicker than taking a cab or a bus and more affordable. $11.55 one way will bring you from Newark Airport to Penn Station.

One-way fare is $13, but you can save by buying roundtrip tickets, as they are $22. You can choose to disembark at Grand Central, Port Authority or Penn Station, The ride takes about an hour. Super Shuttle: Available 24 hours a day, this shuttle provides door to door service for your party. Fares are $15 - $19, depending on destination. The airtrain monorail is the fastest and pretty darn reasonable.

I have never taken a cab but you can easily take a bus from Newark Airport to Port Authority for about $14. The buses run about every 15 minutes from Terminal A and it is a pretty short ride.
