She may back-off from affection - desire sex less often - be out more than usual - have a new wardrobe - be more into healthy activities and diets.
Even if your wife does show some of those signs, it does not necessarily mean she is having an affair. It could have something to do with hormones, or there could be something physcially or mentally wrong.

Is she buying new lingerie that she is not using with you?
Is she paying more attention to her appearance than usual? i.e. Going to the beauty salon when she never did than before.
Is she out of the sudden late from work all the time or taking misterious trips alone for 2-3 hours on a consistent basis?
Does she keep her cellphone conversations secret from you?
Is she on the internet all the time and neglecting you as consequence? Does she close conversation windows as soon as you walk-in the door?
Is she working out when she never worked out before?

Going out shopping and never purchases anything.
She seeming loses interest in sex (but only with you).
Chats online a lot more that she used to.
Her cell phone rings an unfamiliar ringtone and she tells you it's a wrong number - again.
