From the Preamble of the US Constitution (basically like a mission statement for the US Goverment)
At the time that it was written, Enlightenment thinkers believed the legitimate job of government was to protect citizen's lives, basic freedoms, and the ability to do things like earn a living.
"blessings of liberty" would be the rhetoric for basic freedoms like speech and religion. It ties into the rhetoric of being endowed inalienable right from the creator, etc. from the Declaration of Independence, as in blessings from god (creator) as part of inalienable rights.

The blessings of liberty - our freedoms. The freedom to live life the way you want. The freedom so speak your mind without fear of the government locking you away for your opinions. The freedom to live wherever you wish, work as much or as little as you want to maintain your lifestyle.
Securing those blessings means fighting for them against all enemies - foreign and domestic.
