as close. i want to have my best friend be my maid of honor and have my sister be a bridesmaid. do you think this is bad? and does the fact that my fiance is having his brother be his best man change things?

It's your marriage, so you should have full choice over who is in your wedding. Just explain to your sister that you and your friend are so close that you want her to be the maid of honor, but want sis to by a bridesmaid. Your sister should understand. I have three sisters and none were in my wedding, I only had friends stand up with me. There might have been a little grumbling, but years later no one is holding a grudge. My oldest sister recently wed, and I didn't even give a second thought to not being in her wedding. Sometimes friends are closer than relatives, I think we all understands that and I'm sure you're sister will too.

And, no, I don't think it should change anything that your fiance is having his brother be his best man. Like I said, its your wedding, so your decision, just let everyone know your plans tactfully.

Choose exactly who you want and don't worry about them. This is your day, not theirs. Have your best friend be the maid of honor and choose the best man to be the best man. If people throw a tizzy about it, then it proves they're selfish.
