First, snacks and WATER. Airplanes are incredibly dry, for a twelve-hour trip bring a liter, you'll drink it all. In the same vein, bring your favorite chapstick because your lips will dry out. Some people actually bring a small tube of moisturizer with them, I think this is a little over the top, but if you have dry skin anyway you may need it. You should also bring a comfy and warm sweater in case you're cold, and dress in layers in case it's hot. If you are particular about comfort or have neck/back problems, bring a pillow that you know is comforatle when you are sitting up, although most airlines do provide some pillows. If you are on a plane for 12 hours you will almost certainly have movies to watch on the plane, but it might be good to bring a good book, magazine or a couple of crossword puzzles, depending on what you think will keep with occupied. If you have a portable music player it's definitely worth bringing it, because airline radio stations tend to be dull. The last time I was on a plane for 10 hours, the airline gave us all eye masks and earplugs, but I don't know if all airlines do this (it was AirFrance). They were definitely great to have, though, so if you're not sure bring these too!

if it is an international flight, and 12 hours I would guess it is, bring books, iPod, snack food. They will give you plenty of drinks on the flight for free. Also a small pillow is good because the airline pillows suck.

Most of the things people have mentioned, you're going to get anyway on a half-decent airline.
I'd take a spray bottle of water, just to keep your face hydrated, earplugs if you plan to try and sleep and a book in case you don't really like the in-flight movies.

bring your favorite magazines or newspapers. Write letters to friends or family that you haven't written to in a long time. Don't forget stamps to mail them later. On the flight back write postcards out that you forget to do while you are there. maybe thank you notes to people you visited. (of course only if that applies)

A pillow, food, drinks, something to keep you warm ('cause you may not want to use an airline blanket), whatever will keep you entertained (laptop, cell phone, Ipod, book, etc.)
Dramamine is for motion sickness... and yea, stupid friends up mine take it to "trip." It's not healthy at all, unless you have motion sickness.

you have to pack your stuff.. everyone has stuff .. normally when you leave your house you pack your stuff .. stuff is in every ones bags.. stuff has Tobe unpacked and of course when you get off the plane you have to pack your stuff .. To make things more complicated when you get to where you are going unpack your stuff again.. stuff is a big problem.. I could go on but this stuff is getting old...
