Most years, I'd advocate a RB, no question, but this year, if I saw Peyton Manning sitting at #3, I'd snatch him up and high-five myself.
Look at the dropoff in QB's this year compared to RB's. You know there will be one or two other non-RB picks before your 2nd round pick rolls back around. The RB's are very deep this season, by the time you pick in the 3rd round, you will be able to get two RB's that you will be satisfied with.
However, if you go with a RB, you will most certainly not get Manning on the turn, and there's just not that many sure-fire QB prospects this year.
In fact, I've seen some FFB sites (fanball.com being my favorite) advocate waiting to round 3 to pick a RB, because of the depth there, and picking up QB-WR, or even more unconventional, getting your WR in round 1! This again, based on WR's having a big drop off this year. This is not your classic "do the opposite" approach as much as it is reacting to "the market" this season.

Haha, pick my answer because I have had the 3rd pick in every fantasy draft i've had! I used it to draft a running back. There are rules as to what you should do with your first pick and it goes "always take a running back!" It's sensible too because running backs are the most reliable source to get points. Don't get a wide reciever, not matter how much you love Chad Johnson or T.O. because wide recievers are the most inconsistent sources to look for in points. Even if Peyton Manning is available, wait! There are a lot of good quarterbacks out there. In the second round, i'd take another running back because people tend to go for another position, so you'd have a great selection. Now in the middle rounds if you're stuck between taking a 3rd wide receiver or a tight end, take a 3rd or 4th running back because it's better safe than sorry! The rule for taking tight ends early is "if his named ain't Gates, then wait!" I found out the hard way!

Peyton Manning, as long as it's a league where you get 6 points for a touchdown pass. People let star QB's fall way too far in an attempt to follow the traditional wisdom of always drafting the RB first. Thus, RB's end up overvalued and great QB's undervalued in fantasy sports.

Shaun Alexander Larry Johnson LaDainian Tomlinson ... with the number three pick, any of the above should be available.

Since you are at #3, you will not be in a bad position with any of these RBs. I would even look at the players in the order listed ... the following would be my order of importance.

If you're talking about football, take the best out of the remaining 3: Alexander, LJ, or LT. You won't go wrong with either one.
