Two causes actually. The cessation of breathing and the cessation of the heart stopping. Those two, left untreated, will cause certain death within minutes.

We're not immortal, but other than that, there are actually two different reasons people die: apotosis and necrosis. Apotosis is natural death due to age; your body's cells begin to shut down and die as their DNA dictates. Necrosis is death by things like poison/disease; it's not dictated by DNA.
