Easiest test: substitute a compound-form:
for "you and I" -- "we"
for "you and me" -- "us"

Since you'd say 'the difference between US'
you must also say 'the difference between you and ME'.


"Rules":

I/we/he/she/they are subject pronouns (used ONLY for the person(s) doing the action);

me/us/him/her/them are object pronouns.

Prepositions are ALWAYS followed by the object pronoun forms. (This makes sense, since you do not the subject [actor] in a sentence does not come after a preposition.)

Take the other party out and repeat the sentence. Does it sound better as:

'That is the difference between me' or,
'That is the difference between I'

See how it sounds in other similar sentences:

That is the difference for I.
That is the difference for me.

That is the same for I.
That is the same for me.

I would have to say the correct answer is A. 'That is the difference between you and me.'

This is as per basic rules of English grammar. Refer Wren and Martin, the standard text book in Indian schools.

"Me" is correct. To explain simply, if you break the sentence down, "That" is the subject and "difference between you and me" is the object. "I" is a subjective pronoun and "Me" is an objective pronoun. Since it's being used in the object of the sentence, you would use "me."

BETWEEN is a preposition (like over, under, on, with) and is followed by the OBJECTIVE pronoun...me, you, him, her, it, us, them.
