The man that invented the graphic novel is still the best. Will Eisner. His novels are not superhero of even adventure stories, but they compare nicely to the best literature of the 20th century. Read "A Contract With God", "Life On Another Planet", "Invisible People", "To The Heart Of The Storm", and "Minor Miracles" (as well as quite a few others) to see what a graphic novel can be.
Not to take anything away from the titles that the other answers listed, most of them are really good, and should be read, but Eisner was the master. Everybody that came after Eisner is coping him whether they realize it or not (and most comic writers and artist will openly admit that he was the greatest ever). That is why they named the highest award in the comics and graphic novel industry after him.
The best series is probably Cerebus by Dave Sim. It has high points and low points, and should not be read it pieces. It lasted 300 issues (the longest running independent series) and to be appreciated should be read from issue 1 to issue 300. All issues are reprinted in inexpensive trade paper back.

Its such a toss up and it depends on my mood. My first pick is usually Kingdome Come (bought the hardback). I'm actually reading The Watchmen again right now for like the 15th time, adn then there is of course The Dark Knight Returns. I really love all 3, but I gotta give props to my boy Alex Ross, so KC wins.
For a series, hands down 100 Bullets. There will be 100 issues and I beleive its in the 60's right row. Its about various characters, all down on their luck, and are given the opportunity to take their revenge on those responsible for their plight. Its a hardcore crime noir, but it has so much depth and hidden meaning in everything, just like Watchmen. I cannot praise the series, written by Brian Azarello and drawn by Eduard Risso, enough.

Akira by by Katsuhiro Otomo
much as V for Vendetta was a life changing experience...and Dark Knight Returns was a seminal piece of work...I have to go with the eye opening , mind blowing...hair raising experience that was Akira. The anime movie barely scratches the beginning of the story...and that is one seriously good story.
If you liked Rising Stars by Stracynzki then (you had fun with his 'Midnight Nation'.I am sure...) I would suggest Alan Moore's Miracleman...all five graphic novels. As good as Watchmen is...this book is always overlooked.

it's so awesome! So funny yet dramatic! It has an awesome plot and the best characters ever! It gets 9/10! It woulda gotten 100000/10 but the main pairing i don't like.... sigh... what can i do to change it?.... Just write Fan Fics when they do end up together! LOL i'm already doing that!!! Haha! The best series is a tie between The Preacher (by Garth Ennis)and The Sandman ( by Neil Gaiman) !!!!!! I would have to say The Dark Knight Returns by Miller that is the greatest in my opinion MAN !!!!!!
