I have used PHP and MySQL combined with a little bit of JavaScript in LAMP (Linux, Apache, MySQL, PHP) Environment.

PHP and Mysql is the best. It is free and the easiest to use and implement new features and is the best documented language on the web.

I think MySQL/PHP are a better option, because they are FREE!

I don't think there is a best way. You have to start with what you want to accomplish, who your customers will be and what skill set you have.
