the probabilities
1) your mouse needs repair
2) the cable problem
3) the sensors are dirty (Just open the mouse from the bottom see if the sensor's have accumulated dust you can clean it with smoother things)
4) there are viruses which simply makes the mouse non functioning
5) check the port
6) is nothing works just throw the it away and buy a new one its cheaper.

Here's an odd fix for you.
The keyboard seems to work in either port on a PC nowadays, but the mouse only works in 1 of the 2 PS/2 ports.
Try reversing the mouse/keyboard (plug them into each others locations)
Another thing that CAN happen is that there is a pico-fuse on the connector that may have blown. See if the keyboard works in the mouse connector (it should). If it doesn't, the picofuse might be shot and you will need to replace it (if you are a tech, or know one). <--better than replacing the MOBO.

make sure that the mouse cord is plugged in properly at the back of the cabinet and the cord is in healthy position.

if all this thing is fine ,and still your mouse isnt working , then take your mouse to a another system and get it checked there. if its working on the other system then surely something is wrong in your pc , and if its not working on the other pc too then you need to replace your mouse.

your mouse might not be connected or installed properly. check your ports and device manager.

check the usb port (usb port is a rectangular shaped) and change the cable port

plug your mouse cable in your PC properly and restart it again, i think it will solve your problem.

Check whether its plugged in properly, if its usb, try plugging it to another usb...
