I was told that you should always use the parking brake whether you are on an incline or not, because it is better for your car to rest on the brake, as oppossed to resting on the transmission. Any opinions? Will this effect the performance of my transmission?

Yes you should always use the parking brake. The park pawl as rare as it is sometimes do break and it will allow your car to roll away. The pawl is the park pin in your trans, it is only metel, so with stess it can let go and roll away. The 2nd reason to use the parking brake is for the old saying, use it or lose it. The cables are only metal as well and they do rust from mud and rain. So if a long period of time goes by without using them, the cable could possibly rust itself in a none working position and you will fail inspection and have a costly repair on your hands too.

I never use my parking brake unless I am on an incline. I've never had any problems.

You should use your parking brake every few weeks regardless of need just to keep the cable from freezing up.

It doesn't really matter, yes when in park the only think holding your car there is a small pin. But unless another car hits your parked car it isnt going to snap. Always use it on hills but on flat ground (especially your garage) you don't have to worry about it. Make sure that every few months you do pull it and put it back just so it doesnt get rust and get jammed open.

I never use a parking brake on an automatic unless you are on an incline, or are loading it on a trailor to haul somewhere. If you are on a flat, level surface, its not like your putting stress on the transmission anyway.
