I thought the first was James Fenimore Cooper's The Adventure's of Natty Bumpo. But it seems I may have been mistaken as I found this in the article whose link I have put below:

The first American novel, The Power of Sympathy (1789), by William Hill Brown, only shortly preceded the Gothic romance, Wieland (1799), by the first professional American novelist, Charles Brockden Brown.
