That depends upon which movie. In the first one released in 1977 it is definately May the Force Be With You. The Empire Strikes Back in my opinion is debated between the plot twisting line which in the dialogue include both Luke and Vader. Luke:"Obi Wan told me enough he told me you killed my father." Vader: "No I am your father". The other one is right before Han is frozen. Leia' "I love you." Han: "I know." In the Return of the Jedi it is definately "I'll never turn to the darkside. You have failed your Highness, I am a jedi, like my father before me.
The Phantom Menace I could not tell you however in the Attack of the Clones it is most definately when Anakin and Padme are being pulled into the execution arena. "I love you" "I thought you said would destroy our lives" "I think our lives are about to be destroyed anyway and before I die want you to know I truly deeply love you." In the revenge of the Sith it is definately "You were the chosen one. It was said you would destroy the Sith not join them. You were my brother Anakin. I loved you."
According to each film there is one line or set of dialogue that can sum up the entire film. Now just for fun I will iclude my two favorite quotes from LOTR. "I made promise Mr. Frodo, a promise. Don't you lose him Samwise Gamgee, I don't mean to. I don't mean to." Gimli: "I never thought I'd die fighting side by side with an elf." Legolas: "What about side by side with a friend?"

Definitely "May the Force be with you" at about 10 out of 10, with "Luke, I am your father" at about 9.5 of 10.
Once you hear "Force" you know it's Star Wars.
Although there's always the "I have a bad feeling about this" which is only maybe 7 of 10, which has been said in I believe all six episodes.
