As one who has performed in musical productions for quite some time, there are many to consider. Sondheim though is definitely "the chairman of the board". "Sweeny Todd" was groundbreaking, as was "Sunday in the Park with George". Two of my favourites. I also have a place in my heart for classics such as "Oklahoma"(yes Oklahoma...) and "Guys and Dolls".
But if you're talking about the most amazing piece written for the stage, you've got to go with "West Side Story". I toured this show for two years and the music still thrills me to this day. It's social message is just as pertinent now as it ever was and it goes from farce and cutting social satire ("Officer Krupke") to extraordinarily moving lyricism ("Somewhere") in a heartbeat. And nowhere else will you find a more challenging and satisfying musical for an actor, singer or dancer.
Listen to it yourself. You won't be sorry.

Rent was good, I liked the movie better.
Chicago was sexy and fun.
Cats had good music but was a little on the boring side.
Wicked was wonderful but it owes too much to the Wizard of Oz to stand on its own as best.
All the Rodgers and Hammerstein's are classics.
It is difficult to dismiss the Astaire and Rodgers movies.
Of modern epic theater shows, I would say Les Mis. It really draws the emotions out.
From the 60's and 70s' transition era, I would say West Side Story.
And from the earlier romantic/comedy musicals... Singing in the Rain.

MY favorites are rent, wicked, producers, and spamalot
but that doesn't mean they are the best, people are forgetting the classics, like

show boat
but the best one of all times is a very difficult decision
i would say THE PHANTOM OF THE OPERA
it has been on broadway for 18 years and it is still playing
it won 7 tonys in 1988
best musical
best leading actor in a musical
best featured actress in a musical
best director of a musical
best scenic designer
best costume designer
best lighting designer

West Side Story -- hands down.
It stands the test of time, a collaboration of Bernstein and Sondheim and Jerome Robbins.
Based on Romeo and Juliet, it is still timely today in the context of gang warfare and the passions of youth.
Easily the best one of all time! Start with the overture and ride it to the end, it's excellent.

My personal pick would be Les Miserables.
But... it realy depends on how you felt when you first saw or heard it ( insert your pick here). Who was performing and whether the comment the writers were writing about is done in a way and at the right time in your life as to coomunicate with you on a level bordering on your soul
