The title 'Prince of Wales' may be possessed only by the eldest son of a Sovereign.
There is no automatic succession to this title, which at every vacancy becomes merged in the Crown, and is renewed only by the Sovereign.
The title 'Princess of Wales' applies only to the wife of the Prince of Wales. The title cannot be used by the Sovereign's daughter, even if she is heir to the throne. For example, the Queen was known as Princess Elizabeth until she succeeded to the throne.
The present Prince of Wales is 21st in the line, counting several who were never formally invested. He can trace his descent back through the Tudors to the original Princes of Wales, of whom the last native Prince of Wales was Llywelyn ap Gruffydd (1246-82).

Historically England, Scotland and Ireland were Kingdoms, like Spain and Denmark, and they have kings or queens as their head of state. Wales, on the other hand, was a principality, like Monaco or Liechtenstein, and have prince regents as head of state. When the English had finished the invasion of wales the Prince Regent of Wales was deposed and the King of England, Edward I, made his son, (later) Edward II, Prince of Wales. Since then the Eldest son of the Monarch is made Prince of Wales (and they are Heir apparent). If the monarch does not have sons, the eldest daughter will not become princess of wales and the title will be left unfilled.

It's just a title bestowed upon a Prince, the same way Prince Charles the Prince of Wales, the Duke of Cornwall, and several other things. Prince Andrew is the Duke of York, etc. Princess Anne is the Princess Royal, etc. Prince Edward is the Earl of Wessex, etc.
Even Prince Philip, husband of the Queen is the Duke of Edinburgh, among other titles.
