If you have an old computer, go to where you set the screensaver up and then check the box by the screensaver that says to use password and then set your password. Every time the screensaver comes up, you have to enter the password to get back on. This is how you do it if you have windows XP:
control panel
user accounts
change account or change password... I think you can handle it from there.

All the previous answers are right, but one sureshot way of locking ur pc is to set a bios system password, It'll lockdown ur system completely and prevent it from starting unless u hav the passwd...
I havent specified the methodology bcos it is bios dependant,
Just search in google with keywords as Bios and System and Password and "Ur bios company". You'll find the methodology
