I think it is a combo of words meaning many things.
Who opie? Was a common question in a town called Mayberry.
Whoo pie was a major food group in Doctor Suess books.
Put together, it is the sound you make when you feel so good, you click your heals in the air and never come down! You scream, "Whoopie!!!"

It is thought that it refers to sex but it came from a song called Makin' Whoopee. It originally came from a Broadway Musical called Whoopee which debuted on 4 December 1928. The book is by William Anthony McGuire, featuring music by Walter Donaldson and lyrics by Gus Kahn. The original version was produced by Florenz Ziegfeld and starred Eddie Cantor and Ruth Etting. The two-act musical ran for 379 performances. A 1979 Broadway revival of the show ran 204 performances. It was made into a movie in 1930 with Eddie Cantor in the lead role and Betty Grable as one of the Goldwyn girls.

Cantor sang Makin' Whoopee as the centrepiece of the show. It has been covered by everyone from Ella Fitzgerald to Ray Charles. The song as it goes refers to peaks and troughs in a marriage from:

Another bride
Another june
Another sunny
Honeymoon
Another season
Another reason
For Makin' Whoopee.

to the final verse:

He doesn't make much money
Only a five-thousand per
Some judge, who thinks he's funny
Told him he got to pay six to her
He says: "Now judge, suppose I fail."

You'd better keep her,
I think it's cheaper
Than Makin' Whoopee!!"
