Lyndon Johnson became president the moment John Kennedy died. The office of the Presidency is never left vacant. The transition of power is automatic, and the swearing-in ceremony (which for Johnson took place on the plane that would carry JFK's body back to Washington) is a formality.

The Speaker of the House of Representatives becomes next in line for the office, in lieu of an elected Vice President.

He had no VP when he took over after the assassination. When LBJ ran in 1964, Hubert Humphrey was his running mate, so he (obviously) became VP then.
