Aside from the mass floods listed in the Bible and the mass extinctions from the Permian, I would have to say that the worst I've ever witnessed is Hurricane Katrina. There have been numerous earthquakes and natural disasters throughout history, but they didn't wipe out an entire city. By far, the worst disaster the U.S faced that wasn't natural was 9-11.

Science seems to believe that a comet or large meteor hit the earth causing the extinction of the dinosaurs. If that were true then I would have to say that that was the single worst disaster that the earth has ever befallen upon.
If not that then what ever caused the mass extinction of the Dinosaurs would be my vote. What ever it was had to of been pretty traumatic to the Earth.

World War II: millions of Europeans, Japanese and Chinese died. 6 million + Jews and 22 million + Russians died. Countless hundreds of thousands of other nations' citizens died. Except for the US, the economy of the world was nearly shattered.
If you mean natural disaster, then the various outbreaks of plague have been the worst.
