Uh... the creation of the uiniverse? No creation, no human history.
If you are looking for a specific event that involved humans directly, I would have to say the crucifixion of Jesus. Whether you are a believer or not, that one single event changed EVERYTHING. Before then, wars were faught over land, and territory, and domination. After that, many wars were fought for religious reasons. (The Crusades, The Crusades again, The Aztecs and The Spaniards, the current terrorist conflicts- from their side.). The Roman Empire became the Holy Roman Empire. Rulers were deposed and murdered because they were of the wrong faith (Mary, Queen of Scots was killed by Elizabeth I). Entire religions were created because of disagreements with the Church (Henry VIII created the Anglican faith so he could get divorced). Entire cultures were dominated and obliterated in the name of Christ (the American Indians, native tribes in Africa, etc).
And they all came about because one specific individual irritated those in power and disrupted the status quo to the point where they hung him on a slab of wood.

The Greatest Event in Human History would have to be The Battle of Thermopolyea. The greeks should have been easily defeated and forgotten about. Instead, they won an amazing upset victory and went on to invent every great institution and tradition we cherish today. "And the West is history"

The Hygiene revolution, began in the 1800's. One of the earliest pioneers was Herman Lister, yes he invented Listerine. Before the discoveries about hygiene you could die from a paper cut, because people did not understand infections, bacteria, and disease. You could say that Jonas Salk with the Polio vaccine, or any other hundreds of events, but this one event, the hygiene revolution, has saved more lives, and is still saving more human lives than any other discovery before, or since. Hands down.

The day a Semitic man named Abram began worshiping Yahweh and changed his name to Abraham. This led to the formation of the Jewish Religion, then the Christian and Islam. The people who became these religions have dominated most of the world for centuries and have been fighting each other for just as long.

The first time one person discovered God. I say this not because I think everyone needs to be religious--it's just that that discovery lead to so many others. When one feels inspired, it's a spiritual feeling most often. The root of the word inspire comes from that- in spirit.
Inspiration leads to dreams leads to turning points in history.
