Is there any best thing to say when they ask if you've got any questions?

The best way to answer that questions is to be honest: why did you pick that company? Do you like their organization, do you see a way to advance, can you learn a lot, or use a particular skill you have? Want to get other skills from them? These are the reasons to list for wanting that job.

As far as the questions, ask them about their benefit programs, opportunities for advancement or movement within the company, about their community programs or activities, and finally, their assessment of the work culture in the office. You might also ask if you could spend a few hours with the group you'd be working with (upon offer of a job) before accepting.

Best things would probably be to make something up. Say you are very interested in whatever the job involves and tell a load of waffle about how you want to do something positive for the whatever it is. Appear enthusiastic, telling them for example how you really wnt to make a difference to stuff.
The worst things to say are: For the money; I don't want the job, someone else told me I had to apply; Because my benefits will stop if I don't get a job.

Just speak from the heart, but dont get mushy about it. Simply say as I was looking and came upon this job it stood out above the rest and I hope I do the same for you.

That's a common killer question.Just say that "Since I meet all your criterias and since you are looking for special candidates, I am suitable for this".That is why I want this job.
Keep in mind that don't ever become a submissive answerer for that question because it is a question on your POWER QUOTIENT i.e.on your personal awareness.

Good luck.

Be confident:

"I believe I have the capacity to fulfill all criteria for this post, and will do my utmost to be the best person for this job."

Also, be friendly. Don't just answer questions, if you have any queries, ask questions too, it shows you are really interested.
