The traditional heterosexual view of virginity need not apply to gays and lesbians (nor do traditional gender roles!). Losing your virginity does not require the penetration of a penis, in fact it does not require penetration at all. Some people (both straight and gay) have sex and achieve orgasm or feel pleasure without penetration (e.g. tribodisim, S and M, etc.). The definition of when and how ones virginity is lost then, is specific to the individual.

when the hymen (a protective flap of skin that usually stays in tact until a woman experiences vaginal penetration for the fist time) has been broken through a mutual sexual encounter i would imagine. chances are lesbians are not that scientific about it, its probably when they first have lesbian sex involving penetration of some sort.
