About 5-6 hours. Longer than you think, because some of that road is so windy and narrow you have to really slow down. Also, be sure to call the California Auto Club or CHP to ask about road closures, because the main road into Yosemite from the west just got closed by a massive rockslide... so now you have to take extra time to go around to an alternate entrance.

Like the other posters have said, it takes from 4-5 hours, depending on traffic and how many stops you take. I drove it this weekend with two kids and did it in about 4.5 hours. Note that Highway 140 is closed due to rockslides. See (http://www.inciweb.org/incident/announcements/article/236/336/) for the announcement and pictures.
Take Highway 120, instead. The distance is about the same.

it is a good 5 hourish drive depending on traffic, no good freeway all the way there, a lot of two lane highway. Could be longer is you leave during high traffic hours from the Bay area.

About six to eight hours... depending on the time of day you leave the City... traffic on I-5 can be a bear during rush hour!
