My question is really, how to support the patch for the old hole? I know how to cut the new hole .I know that the new hole cut will fit into the old hole, just looking for the best way to support the patch. Is it really that easy that you can just nail wood beams in between
the floor supports and then glue and screw it to the support beams?

first you need to make sure you are cutting a hole in the vent in the new location...figure out if you can get to it from below..then what type of floor are you cutting?..if its just ply wood then its easy to patch but why are you moving it?...and be sure the new location suits the vents needs..vents are either heat or cooling and also can be returns and supplies the location of the vent is prudent to its operation as well...
