Oil immersion objective has got magnification of 100X and used for observing slide which are Gram Stained. Gram staining is nothing but a technique of staining bacteria to confirm their Gram Nature i.e. Gram positive or Gram Negative

When the objective magnification is high (say 100x or more), the lens must be extremely close to the specimen being observed. The immersion oil serves to fill the gap between the two and changes the refractive index allowing the specimen to be seen with more resolution. Make sure you use a cover slip.

Magnifies much larger, with greater resolution. The oil acts as a separate lens, pushing the microbes into a more clear focus.
