Mae West, according to a few sources, said it to Cary Grant in the 1933 film, She Done Him Wrong. One site credits her with the line in the 1970 film, Myra Breckinridge. Most places, though, give her credit as saying it in the 1978 film, Sextette.
Why did she say it? Well, considering in what context the quote (or variations thereof) was used, I'd say the gentleman was, to put it quite succinctly, sporting wood.

The quotation that was made to Cary Grant is, Humph, why don't you come up and see me.
It is funny how some quotes are taken over and have a mind of their own. Mae West was a very lovely lady and she got almost every thing she wanted. Cary Grant was an officer of the law in the picture. In fact it was his very first movie. In the late 1970's she said Oooh, is that a gun in your pocket? Or are you just glad to see me? It wasn't Mae West that said it first. It was a vaudeville comedian but the reason why and who said it is lost in antiquity. Mae West made in popular in 1978 but the saying came most likely from the early 20's.
