Go to control panel, go to system, click the hardware tab, device manager button. Look for anything with a yellow symbol. If it's got one the "Device Drivers" are not loaded for that particular item. Also are you talking about a usb port on the back of the computer or on the front? If it's a front port, you'll want to open up your case and see that the wiring is in fact hooked up for the ports to work. I'd check the ports on the back of the pc to verify the drivers are functioning correctly if this is a front port issue. Then troubleshoot the front ports beginning with verifying that they are actually connected to the motherboard.

1/ When you turn on your computer, tap DEL key or whatever key
you are required to press to enter into the BIOS SETUP. Once you are in the BIOS, go through all the pages carefully, looking for
an entry ONBOARD USB ports - ENABLED/ Dissabled
and make certain that the USB is turned on. SAVE settings and exit
the BIOS.
2/On Re-booting, go into the CONTROL PANEL and SYSTEM /
Device Manager, and see if there is a USB port / HUB / Controller / Root Hub on the bottom of the list, and make certain that there
are no Blue Exclamation marks, Yellow question marks, or
Red marks ( ie. it is working ok ).
If there are no USB devices on the bottom of the list, then
3/ in CONTROL panel, ADD NEW HARDWARE, run through
the automatic Device search, and see if whatever version of windows you have, finds, and sets up the USB, onboard, controller, and ports.

4/ Iff you are referring to an EXTERNAL USB hub device that you are adding to your working, onboard, USB, controller and ports,
then you can have dozens of problems that vary greatly depending on what type of external device you have, whether or not is is externally powered, and what you are plugging it into ( as someone mentioned, front or rear pc ports, etc. ). and which version of OS ( Operating System) you are using ( OS/2, Linux,
TOS, DOS, ME, 2000, NT, 95, 98, XP, UNIX, etc.)

Hope that this gives you a few steps to try.

My friend have a PC with OS Windows 98, and sometime, when he reinstall his PC, it can not detect usb port and usb hub port. What is your OS? Upgrade your OS to Windows Me/2000/Xp if possible.

If the port is on the front, open up the computer, and then check to see that the front panel cable (usually blue or black) is plugged into the system board. Check your motherboard or PC user's manual for more details.
