Best way to spend your life? If you're asking that question, you're doing it. That is, seeking truth. Why are we here? Where will we go? Why is life hard? Why do we want to know why?
What do you see when you look around? Do you see intelligence behind the designs of nature? Or is this the result of a massive explosion. Hmmm... You have an incredible mind, you use only a small portion of it. I've heard about 10 percent. Why is that?
Spend you life seeking, but also, perhaps even more importantly, spend your life loving, or learning to love. All else falls in place.

"highest and best use" is part of Agenda 21 of the United Nations. It is what is happening now in New Orleans with the rebuilding after Hurricane Katrina. It is the Supreme Court decision in Kelo vs. New London, CT that allows a private corporation to take your property for economic development.
First, you have to find your core philosophy. You have to form your own moral compass based on what you think is right. You have to take in all of the information and apply your own filter. Do not blindly accept another's belief without judging its merit yourself. In other words, my core philosophy is to think for yourself and question authority. It is also summarized by the Libertarian "Statement of Principles" as follows:
"We hold that all individuals have the right to exercise sole dominion over their own lives, and have the right to live in whatever manner they choose, so long as they do not forcibly interfere with the equal right of others to live in whatever manner they choose."
Once you know your compass, the rest falls into place and the answer to your question will become clear.

Because life can end so quickly, you have to take every opportunity to cultivate your own happiness, while bearing in mind other people's feelings.
After almost dying when I was 24, I've learned that you need to assess what are the 'big things' versus the 'small stuff' to you. And don't sweat the stuff you can't change, but change what you can.
