If the communication lines have been strained to a point that there is none at all, or a constant line of negative from both sides. One or both decide that there is no time for the other. One is cheating,lying, and show no signs of stopping. Refusal to seek out mediation/help,to find the problem, and or a soulution. So,...if you have lost Faith, Trust, Comfort,while the big pit of total loneliness has taken ahold of you, it's time to cut your losses, and take your lessons in life else where!

lack of interest in each others day to day events,
lack of desire for physical closeness,
lack of nurturance,
lack of maintenance of shared areas.
basically, if you don't feel loved in your partner's presence, and your partner doesn't want to talk about it...that's a huge sign.
