I have tried this route.
Took 170 SBS Bus from Sin to JB Bus Station. (Less than S$2).
Catch a bus from the Bus Station to KL Central. (Less than S$20).
The trick is, at the JB Bus Station (I think it is Lakin), you need to scout for a cheap fair. Many sub-contract sales man.
It is about 4 hrs journey.

Take the express bus. Cheap and convenient. You get to KL in about 5 hours fully refreshed by sl eeping on the coach. Cost around S$25 one way. Return trip is cheaper as you pay in Malaysian ringgit. Total cost less than a tank of gas if you are to drive up. Or fly. Or cycle, or canoe :) LOL!

Take a bus straight to Kuala Lumpur.....u'll enjoy the journey.....it's only about 5 hours...

Realistically speaking (that means we're not talking about walking, cycling crawling etc), if you have a car and more then 1 person going up, driving might be cheaper. Taking one of the express buses should be one of the cheaper methods as well.
Third alternative is to take a fix-ed price taxi up. Again, if there are more then 1 person travelling then it might be more cost efficient by this method.

Take the SBS bus to JB then hop on train to KL cost will be about 10 USD for 1st class ticket. Travel time is about 6 hrs, but the scenery is fantastic movies are shown as well and is not crowded.

FLY AWAY TO THE PLACE THEY CALL KL.Fly up high their twin tower say high to those on the bridge.Ok lah,cycling is the cheapest free is you have your own.I cycle 3 times to kl before.Singapore you go woodlands then,after that,you have to stay on the tip on the highway.I have a gps with me,powerful led red and blue light and spot light to illuminate the road ahead of me.3 led in front 2 behind.Stay safe.

Try taking the bus.
It will get you there in about 5 hours time, the ride is generally comfortable.
Alternatively, take SBS bus across the causeway, then take the Malaysian bus to KL.. that way, you only pay RM from JB to KL.

A 5 hr bus journey. It should cost you between 27 to 40 Sing dollars depending on which provider you choose!

Take the Train, of course.
It's a great way to see the lifestyle of ordinary folk during the train' many stop overs if that's your cup of tea go for it.

Cheapest - bus ( need ~ 5hrs)
Flight - Air Asia ( not sure any schedule from Singpore to KL, but definitely can fly from Senai, Johor to KL)
