Berlen is a spelling variant of Berlin and is a very old German name with origins in Bavaria. You can read a detailed report on it at:

It is most likely German. In olden times, surnames were often descriptive -- Tom, the baker, would be called Tom Baker. Ditto farmer, shepherd, taylor (tailor), etc. In cities, people didn't always have such distinctive occupations, so they were named for the place they came from. Berlen could be a corrupt spelling of Berlin, or there could be a hamlet called Berlen.
