I think we've all been in your situation at one time or another, and can all relate. If you tell him, you risk losing your friendship with you one friend but if you don't' tell him you risk losing the friendship with him if he finds out you knew, not to mention seeing him hurt. So, you're backed into a corner. The whole darned if you do, darned if you do thing.
There's two choices that you have: Go to the friend and tell her what your feelings are, how it's unfair that she lead him on the way that she is and if she continues doing this she's going to hurt him. Tell her that if she doesn't come clean with him or end it then you'll have no choice but to tell him the truth. Try to make her see what she's doing and the consequences of her actions.
The next choice is to try and take yourself out of the situation. Tell her/him you don't want to hear anything about their relationship.
And another choice you have is to talk to him, casually talk about her and see what his feelings are on her. Trying not to be too obvious have him reevaluate their relationship.

I'd ask him If he thinks she really likes him. Then point out that it may not be what he thinks for so and so reason, but then say "I could be wrong". At least you bring up a thought that he can't easily dismiss. Remember you can't make someone change, but you can put ideas into their head. A soft word can break bones.

try talking to the one leading him on, try to persuade her to leave him alone, of course keep it nice. then if that doesn't work you should try to gently convince him to go after someone else, and it sounds like you could be a perfect candidate, but that's just from what i know about the situation.
