That line was never said in the film, but there are two instances where a line similar to that one were uttered:
Ilsa: Play it once, Sam. For old times' sake.
Sam: [lying] I don't know what you mean, Miss Ilsa.
Ilsa: Play it, Sam. Play "As Time Goes By."
Sam: [lying] Oh, I can't remember it, Miss Ilsa. I'm a little rusty on it.
Ilsa: I'll hum it for you. Da-dy-da-dy-da-dum, da-dy-da-dee-da-dum...
[Sam begins playing]
Ilsa: Sing it, Sam.
Sam: [singing] You must remember this / A kiss is still a kiss / A sigh is just a sigh / The fundamental things apply / As time goes by. / And when two lovers woo, / They still say, "I love you" / On that you can rely / No matter what the future brings-...
Rick: [rushing up] Sam, I thought I told you never to play-...
[Sees Ilsa. Sam closes the piano and rolls it away]
And later...
Rick: You know what I want to hear.
Sam: [lying] No, I don't.
Rick: You played it for her, you can play it for me!
Sam: [lying] Well, I don't think I can remember...
Rick: If she can stand it, I can! Play it!
I don't know when people began thinking that "Play it again, Sam" was a quote from the film. That quote was a line in the Marx Bros. film, "A Night In Casablanca" and the title of a Woody Allen play and film.

Ilsa says "Play it once, Sam, for old times' sake"; in response, Sam tries to lie, saying "I don't know what you mean, Miss Ilsa"; and she says "Play it, Sam. Play 'As Time Goes By.' " When Rick hears the song, not realizing yet that Ilsa is there, he rushes up and says "I thought I told you never to play that." Later, alone with Sam, he says "You played it for her and you can play it for me", and then "If she can stand it, I can! Play it!" In A Night in Casablanca, all this dialogue was parodied using the line "Play it again, Sam" — a phrase which has incorrectly become associated with the original film.

Bogart NEVER said "Play it again, Sam." He said many similar things, but not that line. You ought to watch the movie and see for yourself - it's one of the best of all time!

I only saw the movie once and I do speceifically remembering that he did NOT say that verbatim.
He says something like "Play it again, one more time." The piano player/musican's name was Sam and people misquoted it.
