The answer of your question depends somewhat on the particular software you have in mind.
Generally, you need three main kinds of protection - anti-virus, anti-spyware, and a firewall. Anti-virus software protects you mainly against viruses (programs that replicate) and Trojan horses (malicious programs that don't replicate). Anti-spyware software protects you against adware and spyware. Finally, a personal firewall would alert you if something manages to sneak under the other two lines of defense (they usually protect only against malware known to their producer, so a new, unknown malware program has very good chanes to remain undetected) and tries to contact another computer without your knowledge (e.g., in order to allow a remote attacker to control your machine, to install additional malware, to communicate passwords, credit card numbers and other personal data it has gathered, and so on).
Now, you must understand that the lines between these three defenses are not always clear-cut. For instance, many virus scanners also detect a fair amount of adware/spyware. Also, some big commercial anti-virus packages (e.g., McAfee's, Symantec's, etc.) come with a firewall and other goodies.
However, "univeral" products tend to be of worse quality than the "specialized" ones. Think of it this way - a mobile phone with a built-in camera sounds like a good idea, because you don't have to carry both a phone and a camera - but the cameras in these things are usually of much worse quality than a professional digital camera.
So, it is best to use three different products - anti-virus, anti-spyware and firewall.
Now, "which is the best" of these depends very much on your particular needs and priorities. What is more important - price of features? Does it have to be free? Is disinfection important or would you settle just for excellent detection? And so on.
Some good anti-virus programs are Kaspersky Anti-Virus, McAfee's VirusScan and F-PROT.
Of the anti-spyware, I'd recommend Spybot Seach & Destroy (free), although Ad Aware and Microsoft's product are also reasonably good.
Of the personal firewalls, Zone Alarm is very popular but I would advise against it, especially the free version. It causes lots of troubles, constantly causes interruptions of the Internet connection, forgets its settings, and so on. I would recomment using Kerio instead. You can even use it for free; the only things that the registered version has more than the free one are things that you don't really need in a firewall - like web filtering and so on.

Anti-Spy programs: Use more than one. None can catch all the garbage out there: Windows Defender Beta 2, Spybot Search and Destroy, AdAware SE Personal.
This program will work with the AV and AS programs and add an additional layer of protection and it really works great.

Anti-Spam = I don't know examples but there sure are some on the net...
Defragmentator = Windows Defragmenter is slow. Use Diskeeper.
Finally, caution. You may not know that you were tricked by an e-mail, or when you are tricked by buying stuff online.

I just responded to another virus question...my only advice is do not buy over the internet as a download...you have nothing in hand to save yourself if it goes wrong. Go to a store and buy your product there. If it goes wrong you can uninstall and reinstall. Symantec let me down and it cost me dearly.
