if you are maintaining a constant 40mph,, you transmissions lock up torque converter clutch will cut on and off at that speed, it will feel like a bad engine miss, or it could be an overactive EGR valve

It sounds to me like you have a tire that is out of round or has a broken belt inside it. That would be the easiest and least expensive thing to check first. If it is out of round or broken belt, replace the tire and have all of them balanced.
If it is not the tires, you could have lost a balance weight on the torque converter which will make the engine shake very hard and could actually break something inside the engine over time.

Have the wheels balanced first, it doesnt cost that much and it will help even if something else is wrong...
