If you think she's cheating, she probably is. Has she been distant recently? Has her behavior changed? All bad signs, though there could be an innocent answer.
Most people aren't all that good at hiding their activities. Check phone records, especially for cell phones. Look for unusual activity, especially frequent long calls to numbers you don't recognize during non-work hours.
Check credit card statements. Look for purchases that don't make sense, especially at bars, hotels and resturaunts. Pay special attention to time.
Look at bank statements. Look for unusual cash withdrawals from odd ATMs.
Finally, you can ask her flat out. Most people get flustered when confronted this way and won't lie effectively.

Start paying more attention to your wife. Take her out on dates, be sweet to her, do things together that she enjoys. Ask her questions about herself. Get to know your wife, study her. Don't worry if she's cheating. Make sure you are doing what you're supposed to be doing and it will turn out.

Tell her you've something planned all day. Borrow or rent a car and sit down the road and follow her if she leaves. If not "get home early" and check redial on the phone. Or Radio Shack has a simple phone recorder you can attach and listen to the conversations later. It Does Work cheaply and simply. If she is - she'll call.
