John Hancock was the first signer of the Declaration of Independence but was not even at the Constitutional Convention. Neither were Thomas Jefferson, who whote the Declaration, or John Adams.
Many believe George Washington signed first. The thing I like to remember is that because of his poor health, Benjamin Franklin needed help to sign the Constitution. As he did so, tears streamed down his face.
The Constitution was ratified by states in the following order:
Delaware, Pennsylvania, New Jersey, Georgia, Connecticut, Massachusetts, Maryland, South Carolina, New Hampshire, Virginia, and New York. After Washington had been inaugurated, North Carolina and Rhode Island ratified.

Excerpt from the National Park Service's website entitled "Signers of the Constitution."
"Sometime during the afternoon the signing began. All present except Randolph, Mason, and Gerry, or a total of 38 men, affixed their signatures. Accounting for the 39th name, Read signed at the behest of fellow Delawarean Dickinson, who had left the Convention early because of illness. Washington, as the president of the meeting and delegate from Virginia, penned his name first, at the bottom of the fourth page of text. He was followed by the other delegates in the standard congressional voting order, by State generally from north to south: New Hampshire, Massachusetts, Connecticut, Hamilton from New York, New Jersey, Pennsylvania, Delaware, Maryland, Virginia, North Carolina, South Carolina, and Georgia. The names were arranged in two columns, the right-hand one being filled out first."

John Hancock. That's also the biggest signature on the Constitution. That's also why when someone asks you to sign something, they might say, "Put your John Hancock on this."

No one knows. Looking at the constitution there is no way to dtermine who signed first.
