Do you mean a pregnancy test? If so, they work by measuring a substance called HCG or "human chorionic gonadatropin" a substance the body puts out starting shortly after conception.

How do pregnancy tests work?
Am I Pregnant?
Pregnancy tests look for a special hormone in the urine or blood that is only there when a woman is pregnant. This hormone, human chorionic gonadotropin (hCG), can also be called the pregnancy hormone.
The pregnancy hormone, hCG, is made in your body when a fertilized egg implants in the uterus. This usually happens about 6 days after conception. But studies show that the embryo doesn't implant until later in some women. The amount of hCG increases drastically with each passing day you are pregnant.
Many home pregnancy tests claim they can tell if you're pregnant on the day you expect your period. But a recent study shows that most don't give accurate results this early in pregnancy. Waiting one week after a missed period will usually give a more accurate answer

You pee in a cup or on the end of the stick, depending on what kind of test it is, you should pee in a cup first no matter what kind of test you get and dip the end of the stick in the urine, because you might end up ruining the test by peeing on the part that tells you if you are pregnant or not. There are all kinds of tests. I would say that easiest one would be the digital read out that says your pregnant or your not pregnant, that one may be more expensive. The test tests your pregnancy hormones that are in your pee. The urine moves up the stick part of the test to the control part and then to the line that will tell you yes or no. Even a light colored line is positive. There are also directions in the kit when you get it. A blood test from the doctor is always a sure thing.

The strip/part you pee on is kind of like a litmus paper strip that tests for HcG (human chorionic gonadatropin), which is found in urine when you're pregnant.

Read the instructions.
They're usually on a side of the box, or on the little piece of paper you just threw away.
