Based on the recent results of how rookie football players adapted to the NFL - I would say easiest is Running Back and Hardest would be Quarterback or Tight End. Just reviewing the draft and the rookie performances each year you will see the most successful position is the RB. It is really rare to have a rookie player of the year be a QB or TE. Good question.

EASIEST
the O-line is the easiest; as long as you can stand your ground, you can play any of the positions (sans center) no matter what your weight and size is.
on defense, the weak safety is the easiest because you are the last line of defense and mostly you are in a zone coverage, so as long as you cover your man good, QBs (from watching videos) will not usually pass to your position.
HARDEST
defense: the lineback, they have to read the offense, search for holes, watch the play unfold, make quick decisions fast, as well as explode on a snap and with tremendous force; they either blitz , become temporary D-line (when switching formation, say from 3-4 to 5-2), cover man on, or even in passing situations pull out to zone coverage.
offense: the QB, need to read the defense, expect blitzes, stay within the pocket, know when to pass, how far to pass, only person on field regularly who needs to take into account environmental factors all the time (weather, wind, crowd); they are the leader; have a good passing arm, known when and how to throw the ball, know all the plays in the book, even those that the rest of his teammates don't.
OVERALL:
EASIEST: O-tackle
HARDEST: Linebacker

Easiest the HB. Half-Backs can rely on the line, just run the play as designed. Hardiest CB. Try keeping up with some of the NFL recievers.

on defense: the easiest but still very important position is the free safety. you rarely get blocked and you can see everything.
special teams: kicker on kick off
Hardest
Offense: center-you get drilled every play
Defense: Nose tackle-has to scrap in the middle

In high school, WR - you aren't really expected to block and all you have to know is 10 patterns. The RB's have to know the patterns AND the blocking assignments on pass plays. Furthermore, WR's have little contact and can easily take plays off.
Most difficult would be QB fpr obvious reasons.

No position is easy to play, all are vital with vital roles. If one position is a weak link, it will be exposed due to the flexibility and complexity of the game.
