Real hard question even though seems simple. It actually depends on what kind of games you like. And there are so many different types of games. Action, adventure, RPG, sports, simulation, 1st person, 3rd person etc.

There is a very good site named http://gamerankings.com which collects all trustworthy & high quality reviews from different websites and magazines, and then makes the ratio average of it.
According to them, so far, Half Life 2 is the best PC game ever made which secured a ratio of 95.7%. It's a 1st person shooter game. Amazingly enough, the second best pc game is it's predecessor, Half Life. I have Half life 2, and trust me, there is hardly any game which has better graphics & game-play than this. At least 35 "Game of the Year" awards were given to Half-Life 2, which is a record.

The PC version of Resident evil 4 is going to come out in September this year. I think only this game can beat Half Life 2.
I am waiting to see who wins!

Some other very good games, which should not be considered any less than best, are:
1. GTA San Andreas (even GTA 3 and Vice City)
2. Unreal Tournament 2004
3. Civilization 4
4. The Elder Scrolls IV: Oblivion (2006)
5. World of warcraft.
6. Battlefield 2

According to number sales, the SIMS series sold an unprecedented 16 million copies for PC, regarding it one of the best pc games ever.

If you want to play a game for a long time, I suggest GTA San Andreas. It has an unendingly huge world, with over 200 missions accompanied by lots of fun. The variety of missions in here is just amazing.

Battlefied 1942, Battlefield 2. Presentation is outstanding and you will get hours and hours of play. Online, no two games are the same. Highly recommend if you have a PC that'll run them well
