Walk up to the Castle area through the old district- it is a great stroll with lots of nice bars, cafes and shops to look at on the way there and back. I found using a walk that took me from the parlament/embassy area of town towards the castle was unusual and not as crowded - you cross the next bridge along from the signposted one and walk along the river on the other side.

You need to see the zoo, its fantastic! Its a bit out of the way but its cheap (90czk for an adult!) and worth the while (I go there every time I visit Prague!)

For great food and drink i would recommend Jagr's Bar which about half way up wencelas square on the right hand side, its a bit of a pain to find but the food is amazing! Also there is a company called Bohemia Bagel which sells every sort of bagel sandwich you can think of, its also really cheap too! They have 2 cafes, one Masna 2, which is just off the old town square and the other is Ujazd 16 which is across the river across the road from the funicular railway.

Don't forget to buy a travel card from the tourist info desk at the airport for unlimited travel on public transport, they cost from as little as 80czk for 24 hours to 280czk for 7 days, you have to validate this card either in a little yellow machine on the bus or the same type of machine at the train stations, you will use this card more than you think so its really good value for money.

I would avoid the museums as they are all a little bit shoddy, just spend your time roaming around, soaking up the atmosphere and admiring all the beautiful buildings, particularly the Tyn church in the old town square. Trust me when i say your gonna fall in love with the place whatever you see, I first went 7 years ago and I'm due to return for the 17th time next month to exchange contracts on my new house!

If you're fond of a drink....go visit a bar called the Marquis de Sade and buy yourself a pint of Kelt (Czech Guinness). Go on (I think) Thursday or Saturday night and a great singer songwriter named Jamie Marshall will be performing. Terrific atmosphere and a brilliant air of run down decandence (place used to be a notorious brohtel a hundred years ago...). Also try and find your way to Pivovarski Dumm brewery - great beer and wholesome food. The Museum of Sex is hilarious (just off the square by the astrological clock). The funicular up the hill to the park where the Czech's rubbish version of the Eiffel tower is is also a cool way to spend an hour. Enjoy! wish I was going....

The Charles Bridge, Wencleslas Square, the Old Town Square. Drink and food wise, stick to the Czech traditional places - the're miles cheaper than English places.

I agree with the other answers so far, but have to add "The Science museum".... It's brilliant, I spent a day there and still could of spent longer.... look across the river for the huge metronome, The museum is in the park area behind it...

The astronomical clock, obviously. The bridges are nice, too, particularly Karlův most (Charles' Bridge). Generally, try to get a feeling of the atmosphere, and get some decent beer.
