Goto My Computer, right click on the drive for your usb drive, Right Click, and then select FORMAT.

Click on my computer, find the USB drive, double clicks on it. Open the USB drive, select Edit on the top. Move the mouse point to "Select All" to high light all the folders. Click on File and delete them all!

If you're not looking to reformat your drive, simply plug the drive (Memory Stick?) into your computer, open My Computer, and then open the appropriate USB folder and when you're in that folder, select all the files and hit Delete. It'll ask you to confirm, say yes, and the computer will strip the files off that drive.
