Usually something to the effect of:

Tools/Options/Privacy etc....

...where you will find any websites you have blocked. Often it's as simple as clicking on a "remove" button and then restarting the browser.

All depends on the browser you have...

Most people use VPN connections to unblock websites, is good to access blocked sites such as the BBC iPlayer outside of the UK and Hulu out of USA. Also gives anonymous browsing as well which is good when at work I guess.
