Respiration, or volume of the air that our lungs are capable of holding, is measured with a special instrument called sphyrometry (you better check this word, not so sure about spelling!). There is a little tube then you need to put on your lips, then you breed normally and when the nurse tells you "now" you try to breed in as mush air as you can, and then you breed it all out, as fast as you can. The computer measures how much have your lungs expended by breathing, and that's it.
It doesn't hurt at all, actually, it's funny.

Repiration is the movement of air into and out of the lungs. There are a few different values when measuring respiration rate that exist. these are: respiration rate-simply count the number of breaths in one minute (is alway sgood to not let the subject know when you are going to start the counting so they do not change their breathing rate). this rate will vary if the subject is in different positions and during/ after exercise, time of day and mood will also affect rate.
Other repsiration values within a single breath are residual volume (hard to measure as it is the volume of air left in your lungs after you have voluntarly exhaled as much as possible), maximal inspiration volume, the total volume you can suck in after breathing out beforehand, tidal volume which is resting volume of each breath. All of these values can be measured with a flow meter or using douglas bags (big plastic bags that store exhaled air), to give an average over a minute.
The amount of oxygen you use in one minute can be calculated by using douglad bags to collect exahaled air and then analysing the concentraion of nitrogen and carbon dioxide.
Another common test for lung function is the Maximal volume exhaled in 1 second. This tets gives a good indication of the strength of the lung muscles and can help identify if peoples lungs (or actually the muscles around them which help expand and contract the lung volume) aren't strong enough. A flow meter that measures for one second is used for this test.

You can also measure core body temperature using a thermometer in their ear or their back passage to compare repsiration at different levels of activity or after they have eaten a large meal.
