Geralmente eu só vejo a utilizações do DOES em frases interrogativas ou negativas.
Ex: Does she work here ? (I)
Ex: She doesn't work here. (N)

Nas respostas às "tag questions", por exemplo, "Yes, he does",
e ainda é possível quando se quer dar ênfase à afirmativa. Por exemplo:
He does like cats (Ele realmente gosta de gatos, ele gosta mesmo de gatos), He does work well (ele trabalha muito bem).

o verbo TO DO apenas é usado nas expressões interrogativas e/ou negativas (nestas nem em tdas)

é usada sim, para dar ênfase. Como o "indeed".
Ex:
- Does she work here:
- Yes she does work here! And for a long time.
Outro exemplo...
Digamos que vc está com amigos e passa uma garota linda na frente de vcs... aí vc fala..
- Jeez, she's stunning!
Aí um amigo concordando (ratificando) diz...
- Oh my, she does look good.

Pode-se usar o auxiliar em frases afirmativas quando se quer enfatizar o fato.
Ex.: She does love you - Ela realmente ama você.
I do believe you = eu realmente acredito em você.
